Overview

CD19-mbIL15-CAR-T cells is an investigational donor-derived chimeric antigen receptor (CAR) T-cell therapy developed by M.D. Anderson Cancer Center. It is designed to treat patients with relapsed or refractory CD19-positive lymphoid malignancies, such as acute lymphoblastic leukemia (ALL), non-Hodgkin lymphoma (NHL), and chronic lymphocytic leukemia (CLL), particularly following allogeneic hematopoietic stem cell transplantation (allo-HSCT). The T cells are engineered to express a CAR targeting the CD19 antigen, along with a membrane-bound form of interleukin-15 (mbIL15). The mbIL15 component is intended to provide continuous cytokine signaling to the CAR-T cells, enhancing their expansion, persistence, and memory formation in vivo without the toxicity associated with systemic IL-15 administration. The construct also includes a truncated EGFR (HER1t) sequence, which serves as a selection marker and a safety switch that can be targeted by cetuximab to deplete the CAR-T cells in case of severe adverse events.
