**CD19-specific CAR T with dCK knockdown** is an investigational allogeneic anti-CD19 chimeric antigen receptor T-cell therapy engineered to resist fludarabine by suppressing **deoxycytidine kinase** expression with a dCK-directed RNA interference cassette. In the reported preclinical construct, Precision BioSciences used ARCUS gene editing and AAV-mediated targeted insertion to disrupt the endogenous T-cell receptor and insert a transgene encoding a CD19-specific CAR together with the dCK knockdown element, creating a fludarabine-resistant CAR-T product intended to permit concomitant or better-timed lymphodepletion and thereby improve engraftment, expansion, persistence, and antitumor activity. The program appears to have been disclosed as a preclinical platform/product concept rather than a clearly named clinical-stage asset, with proposed relevance to CD19-positive B-cell malignancies such as acute lymphoblastic leukemia and non-Hodgkin lymphoma.
