CD19 specific Chimeric Antigen Receptor T Cell (Nationwide Children's Hospital) is an autologous, genetically modified T-cell therapy designed to target the CD19 antigen expressed on B-cell malignancies. Developed by Nationwide Children's Hospital, this therapy utilizes the Miltenyi Biotec CliniMACS Prodigy system for automated, on-site manufacturing. This approach facilitates a 'fresh' infusion protocol where the CAR-T cells are administered to the patient shortly after the manufacturing process is complete, typically following lymphodepleting chemotherapy with fludarabine and cyclophosphamide. The CAR construct is designed to recognize CD19-positive cells, triggering T-cell activation, expansion, and cytotoxic elimination of malignant B cells. It is primarily being evaluated in pediatric and young adult patients with relapsed or refractory B-cell acute lymphoblastic leukemia (ALL) and non-Hodgkin lymphoma (NHL).
