Overview

This is an autologous cellular immunotherapy consisting of T cells (specifically cytolytic T cells) genetically modified to express a Chimeric Antigen Receptor (CAR) targeting the CD19 antigen. The cells are also engineered to co-express the HyTK fusion protein, which serves as both a selection marker (hygromycin resistance) and a safety suicide gene (thymidine kinase, making cells sensitive to ganciclovir). Developed by City of Hope Medical Center, this therapy is designed to target CD19-positive B-cell malignancies, such as follicular lymphoma. In clinical trials, it is often administered following lymphodepletion (e.g., with fludarabine) and alongside rituximab and interleukin-2 to enhance persistence and efficacy.
