CD19 T cell engager is a preclinical-stage bispecific antibody developed by Nona Biosciences. Built using the company's proprietary heavy chain-only antibody (HCAb) platform (Harbour Mice), the molecule is designed to simultaneously bind to CD19 on B-lineage tumor cells and the CD3 epsilon subunit on T cells. This dual engagement facilitates the formation of an immunological synapse, leading to T-cell activation and the subsequent release of cytotoxic granules, such as perforin and granzymes, to eliminate CD19-positive malignant B cells. The HCAb-based format is intended to provide a smaller molecular size and improved stability compared to traditional IgG-based bispecifics, potentially enhancing tumor penetration and reducing manufacturing complexity. It is currently being investigated for the treatment of various B-cell malignancies.
