CD19 T-cell engager

A CD19 T-cell engager (TCE) is a bispecific or multispecific therapeutic designed to simultaneously bind CD19 on target B cells and CD3 on T cells, thereby redirecting T-cell-mediated cytotoxicity to deplete pathogenic or malignant B cells. Nona Biosciences has utilized its proprietary NonaHCAbFx™ platform, which leverages fully human heavy chain-only antibody (HCAb) Harbour Mice®, to discover and engineer novel CD19-targeting TCEs. Through a function-based screening workflow involving immunization, VH library construction in a CAR lentiviral vector, and high-throughput reporter assays, Nona identified potent CD19-specific TCE leads with diverse epitope binding and superior cytotoxicity compared to reference engagers. This program serves as a platform capability demonstration rather than a formal clinical candidate.
