CD19-targeted allogeneic CAR T cells (specifically referring to the program BMS-986453) is an investigational, off-the-shelf chimeric antigen receptor (CAR) T-cell therapy developed by Bristol Myers Squibb for the treatment of severe, refractory autoimmune diseases. Unlike traditional autologous CAR T therapies that require patient-specific manufacturing from their own cells, this allogeneic approach utilizes T cells from healthy donors, potentially offering a more scalable and rapidly available treatment option. The therapy is designed to target and deplete B cells expressing the CD19 surface antigen, which play a central role in the pathogenesis of various autoimmune conditions by producing autoantibodies and promoting inflammation. By clearing these B cells, the therapy aims to achieve a deep immune reset. It is currently in Phase 1 clinical development for multiple indications, including systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), idiopathic inflammatory myopathy (IIM), and systemic sclerosis (SSc).
