Overview

CD19-targeted chimeric antigen receptor (CAR) T-cell therapy is a revolutionary class of cellular immunotherapy designed to treat B-cell malignancies. The process involves collecting a patient's T cells (autologous) or using donor cells (allogeneic) and genetically engineering them to express a synthetic receptor that specifically targets the CD19 protein, a biomarker found on the surface of nearly all B-lineage cells. Once these 'living drugs' are expanded and infused into the patient, they recognize and kill CD19-positive cancer cells. This platform has led to the approval of several landmark therapies, including tisagenlecleucel (Kymriah), axicabtagene ciloleucel (Yescarta), and lisocabtagene maraleucel (Breyanzi), for indications such as relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL) and various types of non-Hodgkin lymphoma.
