A CD19-targeted T-cell engager is a bispecific immunotherapy designed to redirect effector T cells to eliminate CD19-positive B-cell malignancies. The specific candidate described by IN8bio utilizes a novel gamma-delta (γδ) T-cell engager platform (γδ-TCE) that simultaneously binds to a pan-δ chain sequence on the γδ T-cell receptor and the CD19 antigen on tumor cells. This dual engagement triggers the activation, expansion, and degranulation of both Vδ1+ and Vδ2+ T-cell subsets, leading to the release of cytotoxic molecules such as granzymes, perforin, and pro-inflammatory cytokines like IFN-γ and TNF-α. Unlike conventional αβ T-cell engagers, this platform leverages the innate recognition properties of γδ T cells, including the engagement of stress-associated ligands via receptors like NKG2D and PVR, which may help overcome tumor heterogeneity and MHC restriction in hematologic malignancies such as B-cell acute lymphoblastic leukemia (B-ALL).
