Drug intelligence / Profile preview
CD19.28.40z CAR-T cells is a third-generation chimeric antigen receptor (CAR) T-cell therapy designed to target CD19-expressing B-cell malignancies. Developed by researchers at the Prince of Songkla University, this novel construct incorporates a dual T- and B-cell costimulatory domain consisting of CD28 and CD40 fused to an anti-CD19 single-chain variable fragment (scFv), a CD28 transmembrane domain, and a CD3ζ signaling domain. The integration of the CD40 costimulatory domain, which is typically found in B-cells, alongside the T-cell costimulatory domain CD28, enhances downstream NF-κB signaling (specifically pNF-κB and pRelB) and promotes T-cell stemness and memory signatures. This dual costimulation significantly improves CAR-T cell proliferation, persistence, and long-term tumoricidal activity while reducing T-cell exhaustion and apoptosis, as demonstrated in preclinical models of B-cell lymphoblastic leukemia and B-cell non-Hodgkin lymphoma.
