Drug intelligence / Profile preview
CD19.CAR-SB9 EBVSTs is an armored, allogeneic chimeric antigen receptor (CAR) T-cell therapy being developed by Tikva Allocell for the treatment of systemic lupus erythematosus (SLE). The therapy utilizes allogeneic Epstein-Barr virus-specific T cells (EBVSTs) engineered to express a CD19-targeting CAR, which allows them to selectively deplete pathogenic CD19-positive B cells and plasma cells. To overcome the challenges of allorejection and the requirement for lymphodepletion, the cells are armored with SERPINB9 (SB9), a natural granzyme B inhibitor that enhances resistance to host immune-mediated clearance, and a constitutive IL-7 receptor (C7R) to boost in vivo proliferation and persistence. In preclinical models of SLE, CD19.CAR-SB9 EBVSTs demonstrated robust B-cell ablation and reduced circulating autoantibodies without the need for lymphodepletion.
