Overview

CD19.ENG.iMC T cells are an experimental adoptive cellular immunotherapy consisting of T cells genetically modified to secrete a bispecific T-cell engager (ENG) targeting CD19 and CD3, while simultaneously expressing an inducible co-stimulatory system (iMC). The iMC component comprises a myristoylation-targeting sequence, MyD88 (lacking its TIR domain), the cytoplasmic domain of CD40, and two tandem FKBP12v36 domains, which are activated upon the addition of a small-molecule chemical inducer of dimerization (CID), such as rimiducid. Unlike traditional CAR-T cells, ENG T cells secrete a bispecific molecule that redirects both the engineered T cells and bystander T cells to the target antigen. This platform is designed to provide controlled co-stimulation to enhance T-cell persistence, expansion, and serial killing capabilities in the treatment of CD19-positive hematologic malignancies. In research contexts, these cells often serve as an antigen-specific control for other engager-based constructs.
