Overview

CD19CAR-CD3Zeta-4-1BB Autologous T-Lymphocyte Cells are a form of personalized cell therapy in which a patient’s own T lymphocytes are genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD19 antigen, commonly found on B-cell malignancies. The CAR construct includes an extracellular domain that recognizes CD19, fused to intracellular signaling domains from CD3 zeta (CD3ζ) and the costimulatory molecule 4‑1BB (CD137). This design enhances T-cell activation, proliferation, persistence, and antitumor activity. Upon infusion back into the patient, these modified T cells can recognize and kill malignant B cells expressing CD19. The therapy is primarily investigated for relapsed or refractory B-cell malignancies such as acute lymphoblastic leukemia (ALL), non-Hodgkin lymphoma (NHL), diffuse large B-cell lymphoma (DLBCL), and chronic lymphocytic leukemia (CLL)[2][4][5][7]. The inclusion of 4‑1BB improves CAR-T cell survival and persistence compared to earlier generations[6][8].
