CD19CAR T-cells (University College London) is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy being developed for the treatment of relapsed or refractory primary central nervous system lymphoma (PCNSL). The therapy consists of a patient's own T-cells that have been genetically modified using a viral vector to express a CAR targeting the CD19 antigen, a protein highly expressed on the surface of B-cell lymphomas. In the Phase I CAROUSEL trial, the therapy is administered intravenously following lymphodepleting chemotherapy. For patients with stable or progressive disease, a second dose may be administered intraventricularly via an Ommaya reservoir to bypass the blood-brain barrier and directly target malignant cells within the CNS. The study evaluates the safety, feasibility, and persistence of these cells in patients with this difficult-to-treat condition.
