CD19CAR-TCM cells are an investigational cellular immunotherapy developed by the Fred Hutchinson Cancer Center for the treatment of CD19-positive B-cell malignancies following allogeneic hematopoietic stem cell transplant (HCT). The therapy utilizes donor-derived CD8+ central memory T cells (TCM) that are specifically selected for virus-specificity (typically cytomegalovirus) and genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD19 protein. This specific selection of central memory cells is intended to enhance the persistence and anti-tumor efficacy of the T cells in vivo, while the use of virus-specific donor cells may help mitigate the risk of graft-versus-host disease (GvHD). The primary indications include acute lymphoblastic leukemia (ALL), chronic lymphocytic leukemia (CLL), and various B-cell lymphomas such as diffuse large B-cell lymphoma (DLBCL) and mantle cell lymphoma (MCL).
