Overview

CD19FAP CAR-TEAM is an experimental chimeric antigen receptor (CAR) T-cell therapy designed as a research construct to evaluate the efficacy of dual-targeting strategies in solid tumors. It consists of T cells engineered with a CAR targeting **CD19** and a secreted **T-cell-engaging molecule (TEAM)** that targets **Fibroblast Activation Protein (FAP)** on cancer-associated fibroblasts (CAF) and engages T cells through **CD3**. In preclinical models of pancreatic ductal adenocarcinoma (PDAC), this construct was used as a control to demonstrate that tumor-specific localization (via a tumor antigen like mesothelin) is required for the stroma-modifying effects of the FAP-targeting TEAM to be effective. While CD19 is typically a marker for B-cell malignancies, in this context, it serves as a non-tumor-specific anchor to test the modularity of the CAR-TEAM platform.
