Overview

CD19xCD22 chimeric antigen receptor T-cell therapy is an advanced cell therapy designed to treat B-cell malignancies. It involves genetically engineering a patient's own T-cells to express a chimeric antigen receptor (CAR) that simultaneously recognizes and targets two distinct antigens, CD19 and CD22, which are commonly found on the surface of malignant B-cells. This dual-targeting strategy aims to overcome a significant challenge in CAR T-cell therapy: antigen escape, where cancer cells lose or downregulate the targeted antigen (e.g., CD19) leading to relapse. By targeting both CD19 and CD22, the therapy seeks to enhance the efficacy of tumor cell elimination and reduce the likelihood of relapse, thereby improving patient outcomes in conditions such as relapsed or refractory B-cell acute lymphoblastic leukemia and large B-cell lymphoma. The engineered T-cells, upon encountering cells expressing either or both antigens, become activated, proliferate, and exert cytotoxic effects to destroy the cancer cells.
