Drug intelligence / Profile preview
CD2 x IL2RG bispecific antibody is a preclinical-stage bispecific antibody being developed by Xencor for the treatment of graft-versus-host disease (GVHD) and other autoimmune and inflammatory diseases. The antibody is engineered with blocking arms targeting both CD2 and the interleukin-2 receptor subunit gamma (IL2RG, also known as the common gamma chain or γc). By dual-targeting these pathways, the bispecific antibody selectively blocks IL2RG-mediated γc cytokine signaling (which regulates lymphocyte survival and activation) and CD2-dependent co-stimulation on CD2-positive lymphocytes, such as T cells and natural killer (NK) cells. This dual mechanism provides synergistic modulation of lymphocyte activation, leading to potent suppression of effector activity and prevention of GVHD while minimizing systemic immunosuppression.
