CD20-targeting CAR-T cells with constitutively active gp130 (cGP130) is an experimental cellular immunotherapy designed to treat B-cell malignancies and potentially autoimmune diseases. This construct co-expresses a chimeric antigen receptor (CAR) specific for the CD20 antigen alongside a constitutively active form of the gp130 receptor (cGP130), which mimics IL-27 signaling. The inclusion of cGP130 is intended to drive T-cell proliferation and persistence through the JAK-STAT signaling axis in a cell-intrinsic manner, potentially eliminating the requirement for toxic lymphodepleting chemotherapy (e.g., fludarabine/cyclophosphamide) prior to infusion. Preclinical studies in murine and non-human primate (NHP) models have demonstrated enhanced anti-tumor efficacy and robust expansion without uncontrolled growth or systemic toxicity.
