CD200-IgG1 is a fully humanized monoclonal antibody of the IgG1 isotype that targets CD200, an immunosuppressive molecule overexpressed on acute myeloid leukemia (AML) stem cells. CD200 acts as an immune checkpoint by interacting with its receptor (CD200R) on immune cells, creating an immunosuppressive microenvironment that facilitates leukemic cell survival and immune escape. CD200-IgG1 works by blocking the CD200/CD200R interaction, thereby reversing this suppression and restoring immune cell activity. Additionally, the IgG1 backbone of the antibody enables the induction of antibody-dependent cellular cytotoxicity (ADCC) by NK cells and antibody-dependent cellular phagocytosis (ADCP) by macrophages. Developed by the Oncology Research for Biologics and Immunotherapy Translation (ORBIT) department at the University of Texas MD Anderson Cancer Center, CD200-IgG1 has demonstrated the ability to eliminate CD200-positive AML disease in humanized mouse models.
