Overview

CD22-BBz CAR T cells are a second-generation chimeric antigen receptor (CAR) T-cell therapy targeting the CD22 antigen, a cell surface glycoprotein primarily expressed on B-lineage cells. The 'BBz' designation indicates the inclusion of a 4-1BB (CD137) costimulatory domain and a CD3-zeta signaling domain, an architecture designed to enhance T-cell expansion and long-term persistence compared to first-generation or CD28-based constructs. This therapy is primarily investigated for the treatment of relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL), particularly in patients who have relapsed after CD19-directed CAR-T therapy. Research, such as that conducted at the University of Colorado, explores adjunctive signaling modifications (like ALA-CART) to overcome limitations in targeting CD22-low expressing tumors.
