Overview

CD22 CAR-T is a second-generation chimeric antigen receptor (CAR) T-cell therapy developed by YaKe Biotechnology for the treatment of B-cell malignancies. The therapy involves the genetic modification of autologous T-cells using a lentiviral vector to express a CAR specific for CD22, a surface antigen commonly found on B-lineage cells. The CAR construct includes a CD3ζ activation domain and a 4-1BB (CD137) costimulatory domain, designed to promote T-cell expansion and long-term persistence. It is currently in Phase 1 clinical trials for patients with relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL), chronic lymphocytic leukemia (CLL), and non-Hodgkin lymphoma (NHL). In clinical practice, it is often evaluated as a sequential treatment following CD19-targeted CAR-T therapy to mitigate the risk of CD19-negative relapse and improve durable remission rates.
