Drug intelligence / Profile preview
CD228V (also known as PF-08046031 or SGN-CD228A) is an investigational, second-generation antibody-drug conjugate (ADC) targeting melanotransferrin (CD228, MELTF, or MFI2), a cell-surface glycoprotein highly expressed in multiple solid tumors but with restricted expression in normal tissues. Originally developed by Seagen and subsequently acquired by Pfizer, the asset was licensed to Medicus Pharma in September 2026. CD228V consists of a humanized IgG1 monoclonal antibody (hL49) conjugated to the microtubule-disrupting cytotoxin monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline linker (vedotin platform) with an average drug-to-antibody ratio (DAR) of four. Upon binding to CD228 on tumor cells, the ADC is internalized, releasing MMAE to disrupt microtubule polymerization, induce G2/M cell cycle arrest, and trigger apoptosis. CD228V is being evaluated in a Phase 1 clinical trial (NCT06799533) for the treatment of advanced solid tumors, including melanoma, squamous non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), and esophageal cancer.
