Overview

CD24.28.1XX CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy targeting the CD24 antigen, a "don't eat me" signal expressed on various hematologic malignancies and solid tumors. The construct features a CD28 costimulatory domain and a modified CD3ζ activation domain (1XX), where immunoreceptor tyrosine activation motifs (ITAMs) 2 and 3 are inactivated to calibrate signaling strength. This design aims to enhance anti-tumor efficacy while reducing T-cell differentiation and exhaustion. Preclinical studies at Memorial Sloan Kettering Cancer Center have demonstrated activity against pre-B cell acute lymphoblastic leukemia (ALL), B-cell lymphoma, and acute myeloid leukemia (AML). However, the therapy is associated with significant on-target, off-tumor toxicity, including cytokine release syndrome (CRS) and pulmonary pathology, due to CD24 expression on healthy tissues.
