CD25 CAR-NK92 is an experimental third-generation chimeric antigen receptor (CAR) natural killer (NK) cell therapy targeting the interleukin-2 receptor alpha chain (CD25/IL2RA). Developed by researchers at Inserm and the University of Paris Saclay, the therapy utilizes the NK92 cell line engineered with a CAR construct derived from the scFv of the monoclonal antibody basiliximab. CD25 is overexpressed in chronic myeloid leukemia (CML) during progression to blast crisis and in certain acute leukemias. Preclinical studies demonstrate that CD25 CAR-NK92 cells exhibit specific cytotoxicity, increased degranulation, and interferon-gamma release when exposed to CD25-positive leukemia cells, showing significant anti-leukemic activity in mouse models.
