CD25-depleted autologous peripheral blood mononuclear cells is an investigational cell therapy developed by Providence Health & Services for the treatment of unresectable stage III or stage IV melanoma. The therapy involves the collection of a patient's own peripheral blood mononuclear cells (PBMC) via leukapheresis, followed by the selective depletion of CD25-expressing cells, which are primarily immunosuppressive regulatory T cells (Tregs). The resulting CD25-depleted PBMC population is re-infused into the patient after they have undergone lymphodepleting chemotherapy with cyclophosphamide and fludarabine. This approach is designed to reconstitute the patient's immune system while minimizing the suppressive influence of Tregs, thereby enhancing the efficacy of concurrent treatments such as autologous tumor vaccines and GM-CSF. The primary goal is to stimulate a robust and durable anti-tumor immune response against melanoma cells.
