CD25-depleted donor lymphocytes is an investigational allogeneic cell therapy consisting of peripheral blood mononuclear cells (PBMCs) derived from an HLA-matched donor. The product is selectively depleted of CD4+CD25hiFoxp3+ regulatory T cells (Tregs) using antibody-conjugated magnetic beads, specifically via the CliniMACS CD25 system. By removing these immunosuppressive Tregs, the therapy aims to enhance the proliferation of effector T cells and induce a potent graft-versus-tumor (GVT) immune response in patients who have relapsed after allogeneic hematopoietic stem cell transplantation (HSCT). It has been primarily investigated by the Dana-Farber Cancer Institute for the treatment of relapsed hematologic malignancies, including acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS).
