**CD25 x CTLA-4 DART** is an investigational **bispecific antibody** built on MacroGenics' Dual-Affinity Re-Targeting platform for cancer immunotherapy. The molecule simultaneously targets **CD25** and **CTLA-4** and incorporates an engineered Fc domain designed to enhance binding to activating Fc-gamma RIIIa while reducing binding to inhibitory Fc-gamma RIIb, thereby increasing Fc-mediated effector function and antibody-dependent cellular cytotoxicity. Its intended mechanism is the **selective depletion of tumor-infiltrating regulatory T cells** that co-express CD25 and CTLA-4, with the goal of reducing immunosuppression in the tumor microenvironment while sparing antitumor effector T cells. Available public information is preclinical, including AACR 2020 data showing Fc-dependent depletion of activated FoxP3-positive Tregs with relatively limited effects on activated FoxP3-negative effector cells; no distinct proprietary non-code INN or brand name has been publicly disclosed.
