Overview

CD276 binder CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target CD276 (also known as B7-H3), an immune checkpoint molecule overexpressed in various solid tumors, including glioblastoma. Unlike conventional CAR T cells that use single-chain variable fragments (scFv) for antigen recognition, this therapy utilizes a de novo computationally designed protein binder. These binders are engineered using advanced computational tools such as Rosetta FastDesign and AlphaFold2 to optimize affinity and stability. This approach is intended to enhance the durability of CAR expression on the T-cell surface and improve resistance to the immunosuppressive tumor microenvironment, potentially leading to more effective and sustained antitumor responses in solid malignancies.
