Overview

CD276-CAR-T cell is a type of Chimeric Antigen Receptor (CAR) T-cell therapy designed to target and eliminate cancer cells expressing CD276, also known as B7-H3. B7-H3 is a member of the B7 family of immune checkpoint molecules, often overexpressed on various solid tumor cells, making it a compelling therapeutic target. The CAR construct typically incorporates CD276-binding moieties (e.g., scFv or nanobody) linked to signaling domains like 4-1BB and CD3ζ, enabling T cells to recognize and kill CD276-positive tumor cells. A notable challenge with CD276-CAR-T cells is the upregulation of endogenous CD276 on the CAR-T cells themselves upon activation, which can lead to fratricide (self-killing) and limit their expansion and persistence. Research indicates that CRISPR-mediated knockout of endogenous CD276 in the CAR-T cells can mitigate this issue, enhancing proliferation and cytotoxicity.
