Overview

CD276 CAR T cells are a form of chimeric antigen receptor (CAR) T cell therapy engineered to target the tumor-associated antigen CD276, also known as B7-H3. CD276 is an immune checkpoint molecule highly expressed on a wide range of solid tumors and hematologic malignancies but with limited expression in normal tissues, making it an attractive immunotherapy target[1][3][5]. The mechanism of action involves genetically modifying patient-derived or donor-derived T lymphocytes to express a synthetic receptor that recognizes and binds to the extracellular domain of CD276 on tumor cells. Upon binding, these engineered T cells become activated and mediate cytotoxicity against the cancer cell through cytokine release and direct killing[1][4][5]. Preclinical studies have demonstrated potent anti-tumor activity in models such as rhabdomyosarcoma (RMS) and acute myeloid leukemia (AML), with efficacy correlating to the level of CD276 expression on tumor targets[1][5]. This approach is being explored for various cancers including RMS, AML, esophageal squamous cell carcinoma (ESCC), and other solid tumors[2][4][6].
