CD276-targeted CAR-T cells (also known as B7-H3-targeted CAR-T cells) represent an investigational cellular immunotherapy platform consisting of T cells genetically engineered to express a chimeric antigen receptor (CAR) specific for CD276 (B7-H3). CD276 is an immune checkpoint molecule belonging to the B7 family that is frequently overexpressed across various solid tumors—including lung, esophageal, and colorectal cancers—while maintaining restricted expression in normal tissues. Upon binding to CD276 on the surface of malignant cells, these CAR-T cells undergo activation and proliferation, mediating targeted tumor cell lysis. This therapeutic approach is being evaluated in early-phase clinical trials, including a Phase I study sponsored by The First Affiliated Hospital of Zhengzhou University to assess safety and preliminary antitumor efficacy in patients with advanced solid tumors.
