CD28 x B7H3 bsAb is a bispecific antibody developed by WuXi Biologics designed to enhance T-cell activation and anti-tumor activity. It functions by providing a costimulatory signal (Signal 2) to T cells through agonistic binding to CD28, while simultaneously targeting B7H3 on tumor cells. This dual engagement aims to improve T-cell proliferation, survival, and response durability, particularly when combined with anti-PD-1 therapy or CD3 T-cell engagers. Preclinical studies have demonstrated its ability to increase IL-2 release, induce strong killing of tumor cells in vitro and in vivo, and provide anti-apoptotic signals to T cells, suggesting its potential for treating a broad range of solid tumors.
