Overview

CD3ɛ shRNA is a short hairpin RNA (shRNA) designed to silence the expression of the CD3 epsilon (CD3ɛ) subunit of the T-cell receptor (TCR) complex through RNA interference. In preclinical research, particularly involving ataxia-telangiectasia mutated (ATM) deficient models, it is utilized to investigate the dependency of thymic T-cell lymphomas on pre-TCR signaling. By knocking down CD3ɛ, the assembly and constitutive signaling of the pre-TCR are inhibited, which has been shown to significantly impair the survival of surface CD3ɛ-high (sRhi) lymphoma cells. This suggests that the pre-TCR signaling pathway is a critical survival requirement for certain subsets of T-cell acute lymphoblastic leukemia (T-ALL) and represents a potential target for clinical intervention.
