Overview

CD3 x B7-H3 BiTE refers to a class of bispecific T-cell engager (BiTE) antibody constructs designed to simultaneously target CD3 on T cells and B7-H3 (CD276) on tumor cells. B7-H3 is a transmembrane protein highly expressed in various solid tumors—including head and neck squamous cell carcinoma (HNSCC), glioblastoma, and ovarian cancer—but has limited expression in normal tissues. By physically bridging cytotoxic T cells to B7-H3-expressing cancer cells, these biologics redirect T-cell killing activity and bypass traditional MHC-restricted recognition. Several versions have been developed, such as MacroGenics' MGD009 (orlotamab) and the ExMab-based EX105, and are primarily in preclinical or early clinical development for the treatment of advanced solid malignancies.
