This investigational cell therapy, developed by Baylor College of Medicine, consists of autologous Epstein-Barr virus (EBV)-specific cytotoxic T-lymphocytes (CTLs) genetically modified to express a chimeric antigen receptor (CAR) targeting CD30. Designed for the treatment of relapsed or refractory CD30-positive Hodgkin's lymphoma and non-Hodgkin's lymphoma, the therapy employs a dual-targeting mechanism: the native T-cell receptors (TCRs) recognize EBV antigens, which enhances the persistence of the cells in vivo, while the CD30-specific CAR enables the direct killing of CD30-expressing malignant cells, such as Hodgkin Reed-Sternberg cells. This approach aims to overcome the limitations of standard CAR-T therapies by leveraging the longevity of virus-specific memory T cells.
