Overview

CD317-CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CD317 (also known as Bone Marrow Stromal Cell Antigen 2, BST2, or Tetherin), a transmembrane protein overexpressed in glioblastoma and other malignancies. The therapy involves engineering T cells to express a CAR specific to CD317, enabling them to recognize and eliminate tumor cells. To optimize efficacy and prevent fratricide (self-killing of engineered T cells that may express CD317), researchers have employed techniques such as RNA interference or CRISPR/Cas9 to silence or knockout CD317 expression in the T cells themselves. Preclinical studies in orthotopic glioma mouse models have demonstrated significant anti-tumor activity and prolonged survival.
