Drug intelligence / Profile preview
CD317-targeting CAR T cells are a preclinical-stage chimeric antigen receptor (CAR) T-cell therapy designed to target the transmembrane protein CD317 (also known as bone marrow stromal antigen 2, BST-2, or tetherin). CD317 is highly and stably expressed in glioblastoma and other solid tumors, while showing minimal expression in healthy brain tissue, making it a promising immunotherapeutic target. To overcome target-driven fratricide caused by CD317 expression on T cells themselves, these second-generation CAR T cells are engineered to co-express a specific short hairpin RNA (shRNA) that silences CD317 expression in the transduced T cells. This modification significantly enhances the viability, proliferation, and cytolytic activity of the CAR T cells. Developed by researchers at the University Hospital Zurich, the University of Zurich, and Christian-Albrechts-University Kiel, this therapy has demonstrated potent anti-glioma activity and prolonged survival in orthotopic xenograft mouse models of glioblastoma.
