CD31xGPVI bispecific antibody is a first-in-class preclinical-stage bispecific antibody being developed by Tridek-One (a spin-off from INSERM) for the treatment of thromboinflammatory cardiovascular diseases, including ST-segment elevation myocardial infarction (STEMI), stroke, peripheral artery disease, and abdominal aortic aneurysm. Designed in an Fc-silent format, the antibody dual-targets the inhibitory immunoreceptor tyrosine-based inhibitory motif (ITIM)-containing receptor CD31 (PECAM-1) and the immunoreceptor tyrosine-based activation motif (ITAM)-containing platelet collagen receptor glycoprotein VI (GPVI). By agonizing CD31 and blocking GPVI, the therapeutic candidate inhibits collagen-driven platelet activation and aggregation, prevents platelet-leukocyte aggregate formation, and reduces NETosis, thereby providing both antithrombotic and anti-inflammatory efficacy while minimizing bleeding risks.
