Overview

CD32b CAR-T is an investigational autologous chimeric antigen receptor T cell therapy engineered to express a CAR targeting CD32b (also known as FcγRIIb), an inhibitory Fc receptor highly expressed on malignant B cells, especially in chronic lymphocytic leukemia (CLL) and other B-cell lymphomas. The mechanism of action involves the specific recognition and cytotoxic elimination of CD32b-expressing B cells by engineered T cells. Preclinical studies demonstrated effective lysis of CD32b-positive leukemic cell lines and primary CLL cells in vitro and in xenograft models. CD32b is a promising target due to its homogenous and high-density expression on CLL cells, with potential benefits for patients who are resistant to existing CD19 CAR-T therapies. The therapy remains investigational, with ongoing research into optimal safety modifications to reduce off-target toxicity[1][3].
