Overview

CD33-28z CAR T is a second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target CD33 (also known as Siglec-3), a transmembrane protein highly expressed on the surface of leukemic blasts in the majority of patients with acute myeloid leukemia (AML). The CAR construct incorporates an anti-CD33 binding domain (typically an scFv) linked to a CD28 costimulatory domain and a CD3ζ (zeta) signaling domain. This specific architecture is intended to provide both primary activation and costimulatory signals to the T cell upon antigen engagement. In preclinical studies, CD33-28z CAR T cells have demonstrated potent anti-leukemic activity and rapid expansion, although they are often used as a benchmark to evaluate newer CAR platforms aimed at improving metabolic resiliency and long-term persistence, such as the ALA-CART platform developed at the University of Colorado Anschutz Medical Campus.
