Overview

CD33 4-1BB CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target CD33 (Siglec-3), a transmembrane protein frequently expressed on the surface of leukemic blasts in acute myeloid leukemia (AML). This specific CAR construct incorporates a 4-1BB (CD137) co-stimulatory domain, which is intended to enhance the metabolic fitness and long-term persistence of the T cells compared to first-generation or CD28-based CARs. Research conducted by the National Institutes of Health (NIH) and other institutions has evaluated these cells in preclinical models, noting that while 4-1BB domains may offer superior persistence, they might exhibit lower immediate potency and activation levels than CD28-based constructs in certain AML contexts, potentially influencing the emergence of extramedullary disease. The therapy is primarily developed for the treatment of relapsed or refractory AML.
