CD33 ALA-CART is an experimental chimeric antigen receptor (CAR) T-cell therapy designed for the treatment of acute myeloid leukemia (AML). Developed at the University of Colorado Anschutz Medical Campus, the ALA-CART (Adjunctive LAT Activating CAR T) platform utilizes a bicistronic construct that expresses both a standard second-generation CAR targeting CD33 and an adjunctive CAR containing the intracellular signaling domain of the Linker for activation of T cells (LAT). This design is intended to overcome signaling deficits inherent in standard CAR T cells, specifically the inefficient activation of the LAT scaffolding protein in response to low antigen levels. By restoring LAT signaling, CD33 ALA-CART cells demonstrate enhanced in vivo potency, increased persistence, and an enrichment of T stem cell memory cells compared to traditional CD33-directed CAR T cells in preclinical models.
