CD33-AlloCART is an allogeneic chimeric antigen receptor (CAR) T cell therapy under development as a potential treatment for acute myeloid leukemia (AML). This therapy utilizes non-viral engineering methods, specifically MaxCyte's ExPERT™ electroporation platform, for the highly efficient delivery of biomolecules. The process involves the use of Sleeping Beauty 100X transposase mRNA and CD33-CAR transposon minicircle DNA to introduce the CAR, alongside CRISPR-Cas editing targeting the TRAC and B2M loci. This approach aims to reduce immunogenicity and manufacturing costs while preserving T cell viability and functionality, enabling the generation of functional CD33-AlloCART cells at a clinically relevant scale.
