CD33 antibodies are a class of monoclonal antibodies designed to target the CD33 antigen (Siglec-3), a transmembrane protein primarily expressed on the surface of myeloid lineage cells, including myeloid leukemia blasts. These antibodies are utilized in various therapeutic formats, including unconjugated antibodies that induce antibody-dependent cellular cytotoxicity (ADCC), antibody-drug conjugates (ADCs) like gemtuzumab ozogamicin, and as the binding domains for bispecific antibodies or chimeric antigen receptor (CAR) therapies. Research, such as that presented at ASH 2024, highlights the importance of epitope selection, suggesting that targeting the membrane-proximal C2-set domain rather than the distal V-set domain may enhance the efficacy of NK cell-mediated and T-cell-mediated immunotherapies by reducing the distance between the effector and target cells.
