CD33 BBz CAR-T cells are a type of chimeric antigen receptor (CAR) T-cell therapy, a form of adoptive immunotherapy. These T cells are genetically engineered to express a CAR that specifically recognizes and binds to the CD33 antigen, which is highly expressed on the surface of acute myeloid leukemia (AML) blasts and some normal myeloid precursors. The 'BBz' in the name indicates the inclusion of 4-1BB (CD137) and CD3 zeta (CD3ζ) co-stimulatory domains within the CAR construct. Upon binding to CD33-expressing cancer cells, the CAR activates the T cells, leading to their proliferation, secretion of pro-inflammatory cytokines (such as IL-2, IFN-γ, and TNF-α), and direct cytotoxicity through the release of perforin and granzyme, ultimately resulting in the destruction of the targeted cancer cells. This therapy is under investigation for the treatment of relapsed/refractory AML and other CD33-positive hematological malignancies.
