Overview

CD33 CAR-T is a type of chimeric antigen receptor T cell therapy that targets the CD33 antigen expressed on acute myeloid leukemia (AML) cells. This immunotherapy involves genetically modifying T cells to express a receptor that recognizes CD33, enabling them to bind to and destroy CD33-positive cancer cells. Various designs have been developed, including membrane-proximal CD33-IgC-targeting CARs that show improved efficacy compared to membrane-distal targeting versions, and drug-regulated systems like DARIC33 that can be controlled with rapamycin administration. CD33 CAR-T therapy has demonstrated significant cytotoxicity against AML cell lines and primary samples, even at low effector-to-target ratios, making it potentially effective in high tumor burden settings.
