Overview

CD33 CAR T cells are a type of chimeric antigen receptor (CAR) T-cell therapy engineered to target the CD33 antigen, primarily investigated for the treatment of relapsed or refractory acute myeloid leukemia (AML). The specific version discussed in the provided context is functionally enhanced by the addition of a potentiating molecule linked to human CD33 scFv by P2A. This therapy aims to eliminate CD33-positive leukemic cells. Early clinical data from a Phase 1 study (NCT04835519) in r/r AML patients showed promising safety and efficacy, with some patients achieving complete remission with incomplete hematologic recovery (CRi) and remaining disease-free for extended periods. However, side effects such as cytokine release syndrome (CRS), neurotoxicity, and depletion of normal CD33-positive cells were observed.
