CD33-CD70 CAR T cells are an investigational cell therapy approach designed to treat acute myeloid leukemia (AML) by simultaneously targeting two antigens, CD33 and CD70, which are frequently expressed on leukemic blasts. This dual-targeting strategy aims to overcome the challenges of antigen heterogeneity and antigen escape, which often lead to relapse in single-target CAR T-cell therapies. Different architectural approaches are being explored, including "co-transduced" CAR T cells that express chimeric antigen receptors for both targets on a single cell, "pooled" approaches combining two single-target CAR T populations, and CAR T cells targeting one antigen (e.g., CD70) while secreting bispecific T-cell engaging antibody molecules (TEAMs) against the second (e.g., CD33). Research is being conducted by institutions such as Massachusetts General Hospital and Emory University, with preclinical studies demonstrating enhanced cytotoxicity and the ability to eradicate tumors with heterogeneous antigen expression.
