Overview

CD33-TCB is a T-cell engaging bispecific antibody (TCE) designed to target the CD33 antigen, a transmembrane receptor frequently expressed on the surface of leukemic blasts in acute myeloid leukemia (AML). Developed using Roche's proprietary CrossMAb technology, CD33-TCB typically utilizes a 2+1 molecular format, which features two antigen-binding domains for CD33 and one for the CD3 epsilon subunit of the T-cell receptor complex. This configuration facilitates the recruitment and activation of cytotoxic T cells directly to CD33-positive tumor cells, bypassing traditional MHC-mediated antigen presentation. While CD33-TCB has demonstrated potent anti-leukemic activity in preclinical models, its clinical utility has been limited by on-target, off-tumor toxicity against healthy hematopoietic stem and progenitor cells (HSPCs), which also express CD33, leading to severe myelosuppression and impaired hematological recovery.
