CD33 V-set CAR T cells are a chimeric antigen receptor (CAR) T-cell therapy designed to target the membrane-distal V-set domain of the CD33 antigen, which is a well-established marker for acute myeloid leukemia (AML). These CAR T cells typically utilize single-chain variable fragments (scFvs) derived from antibodies such as hP67.6 (My96). While this approach has been evaluated in clinical settings, research indicates that targeting the V-set domain, which is further from the cell membrane, may result in less potent effector functions compared to targeting membrane-proximal domains. These cells are primarily investigated for the treatment of AML and other CD33-positive hematologic malignancies.
